ABSTRACT

In recent years, the MYB-related gene family has been found pivotal in plant growth and development. MYB-related gene family in Angelica dahurica var. formosana was systematically investigated based on "Chuanzhi No. 2" through transcriptome database search and bioinformatics and the temporal and spatial expression patterns were analyzed through real-time fluorescence-based quantitative polymerase chain reaction(PCR). The results showed that 122 MYB-related proteins family were identified, mainly including the unstable hydrophilic proteins with good thermal stability. Most of the proteins were located in nuclei. The majority of the proteins had the structures of random coil and α-helix. Five MYB-related proteins family of A. dahurica var. formosana had membrane-binding domains. The conserved domain analysis of MYB-related proteins family of A. dahurica var. formosana showed that the MYB domains of genes in five subgroups, similar to 2 R-, 3 R-, and 4 R-MYB proteins, contained three evenly distributed Trp(W) residues in the MYB repeat sequence. The phylogenetic analysis of MYB-related proteins family in A. dahurica var. formosana and Arabidopsis thaliana showed that the MYB-related members were unevenly distributed in five subgroups, and A. thaliana and A. dahurica var. formosana had almost the same number of genes in the CCA1-like subgroup. There were differences in the number, type, and distribution of motifs contained in 122 encoded proteins. Transcription factors with similar branches had similar domains and motifs. The expression pattern analysis showed that the transcription factors AdMYB53, AdMYB83, and AdMYB89 responded to hormones to varying degrees, and they were highly expressed in leaves and responded quickly in roots. This study lays a foundation for further investigating the function of MYB-related transcription factors of A. dahurica var. formosana and solving the corresponding biological problems such as bolting early.

ABSTRACT

Objective The purpose of this study was to investigate the differential expression of microRNA in gefitinib acquired resistant lung cancer cells and non-resistant cells and their roles on cell growth.MethodsGefitinib resistant NSCLC A549 and H1299 cells were constructed. The cell activity was detected by CCK-8 assays. Real-time PCR was used to detect microRNA expression. The intracellular microRNA expression profile was detected by microarray. The transfection of miRNA-7 mimic was used to up-regulate the expression of mir-7. EGFR protein expression was detected by Western -blotting.ResultsThe IC50 value of gefitinib resistant cells was 3 fold than that of their parental cells. The drug-resistant cells grew slowly compared with their parental cells. 43 miRNA were significantly changed in the gefitinib resistant cells, including 25 up-regulated miRNA and 18 down-regulated miRNA. The IC50 was significantly decreased and the EGFR expression was negatively regulated after up-regulated of mir-7.ConclusionmiR-7 expression negatively regulates EGFR expression and decreased expression of miR-7 is one of the mechanisms leading to gefitinib resistance in non-small cell lung cancer cells.

ABSTRACT

Background Researches showed that elevatory blood glucose level results in long-term damage of cells and tissue,or metabolic memory phenomenon, and manipulation of hyperglycemic memory is a good approach in the prevention of diabetic complications. However, its mechanism is not clear. It is speculated that the pathogenesis of diabetic retinopathy (DR) in diabetic patients may be associated to related mechanisms.Uncoupling proteins (UCPs) can decrease the production of reactive oxygen species (ROS),which may be related to DR.Objective This study was to explore the association between DR and the single nucleotide polymorphisms (SNPs) of UCP genes in Chinese Han population with type 2 diabetes. Methods A cross-sectional study was performed. This study was approved by Ethic Committee of Affiliated First Hospital of Shanghai Jiao Tong University and complied with Declaration of Helsinki, and written informed consent was obtained from each subject prior to any medical examination. One thousand eight hundreds and seventy-five patients with type 2 diabetes mellitus were enrolled in Xinjing district of Shanghai city by cluster sampling from November 2014 to January 2015.The demographic and medical baseline characteristics, ocular examination and laboratory tests were obtained and periphery blood of 2 ml was collected for extraction of DNA.Eight tag SNPs of UCP1,three tag SNPs of UCP2,and seven tag SNPs of UCP3 were selected as marker locus for the detection of genotype by Sequenom Mass ARRAY.Matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) mass spectrometry platform were used for genotyping.Hardy-Weinberg equilibrium (HWE) analysis, allele and genotype frequencies, haplotype analysis, and association tests for DR and SNPs were performed by SAS and SHEsis software. Results A total of 530 DR patients were checked out from 1 875 subjects with type 2 diabetes mellitus, with the detection rate of 28.27%.rs660339 locn of UCP2 gene and rs1626521,rs668514 locus of UCP3 gene appeared to have low detectable rates, and the secondary allele base frequency of rs632862 in UCP2 gene was <0.01 and rs15763 of UCP3 gene was unmatched with HWE,therefore, these locus analysis was not included. In 13 SNPs locus included in the analysis, only 2 SNPs of UCP1 gene were related to DR.Compared with the non-diabetic retinopathy (NDR) patients, the G allele frequency of rs10011540 was increased (P =0.03,OR =1.31,95 % confidence interval[CI] =1.03-1.67,and T allele frequency of rs3811787 was decreased (P=0.04,OR=0.86,95% CI=0.75-0.99) in DR patients.Genotyping detection showed that the C/C and A/A frequencies of rs3811790 in UCP1 gene were significantly more and C/A frequency was less in DR patients than those in NDR patients (all at P<0.01).The logistic regression analysis indicated an association of SNPs of rs10011540 and rs3811787 with DR independent from glucose and disease duration. Conclusions The SNPs of rs10011540 and rs3811787 locus in UCP1 gene are associated with DR in Chinese type 2 diabetes patients.

ABSTRACT

Objective To observe the serum adiponectin, leptin level, and adiponectin/leptin ratio (A/L ratio) in elderly male patients with normal BMI and visceral adipose deposit but without metabolic syndrome. Methods A total of 109 elderly males (≥60 years old) were enrolled whose BMI were all less than 25 kg m and without metabolic syndrome. They were divided into non visceral adipose deposit group (n = 67) and visceral adipose deposit group (n = 42 ). Serum levels of adiponectin and leptin were measured by radiological immunological assay (RIA) and the A/L ratio was calculated. Visceral adipose deposit was defined as visceral adipose area more than 100 cm2.Metabolic syndrome diagnosis was according to the definition provided by China Diabetes Society (CDS) in 2004. Results (1)The visceral adipose area [(135.56±31.72)cm2 vs. (68. 65±22.64)cm2 , P< 0. 001] and BMI [(22.94 ± 1.35 ) kg/m2 vs. ( 21.38 ± 2.55 ) kg/m2 , P < 0.001] were significantly higher in visceral adipose deposit group than in non visceral adipose deposit group. The A/L ratio was significantly lower in visceral adipose deposit group than in non visceral adipose deposit group ( 2. 17 ± 1.77 vs. 4.54 ± 7.00, P = 0. 034 ) while there was no difference in serum adiponectin or leptin level between those two groups. (2)The A/L ratio was negatively correlated with BMI (r=-0.552, P<0.001), waist circumstances (r=-0.390, P<0.001) and abdominal visceral fat area (r=-0.341, P<0. 001 ). Conclusions The A/L ratio decreases in elderly men with normal BMI and visceral adipose deposit. A/L ratio could be useful in scanning those patients with visceral adipose deposit whose BMI is normal among elderly men.

ABSTRACT

The effects of insulin or insulin in combination with chemotherapeutic drugs on the proliferation and apoptosis of endometrial carcinoma cells were examined with an aim to determine the efficacy and safety of insulin in endometrial cancer therapy. Ishikawa and Hec-1A cells were treated with insulin and/or paclitaxel. Cell proliferation was assessed by MTT assay. Cell cycle and cell apoptosis were determined by flow cytometry (FCM). Survivin gene expression was detected by RT-PCR. Our results showed that in a certain range of working concentrations and action time, insulin could mildly augment cell proliferation and the percentage of S phase cells in endometrial cancer (Ishikawa/Hec-1A) cells. Insulin plus paclitaxel (combination group) could significantly inhibit cell proliferation (69.38%±2.32% vs 40.31%±4.52% with Ishikawa; 64.11%±6.33% vs 45.89%±3.27% with Hec-1A) and increase cell apoptosis compared with treatment with paclitaxel alone (paclitaxel group). Survivin gene expression was also significantly decreased in combination group as compared with paclitaxel group. We are led to conclude that insulin can mildly augment cell proliferation and present chemotherapy sensitivity in endometrial cancer cells. Insulin can be to used safely and efficiently in endometrial cancer therapy.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the role of monitoring sex chromosome chimeric status by fluorescence in situ hybridization (FISH) in the identification of leukemic extramedullary relapse and post-transplant lymphoproliferative disease (PTLD) in acute lymphocytic leukemia (ALL) after allogeneic hematopoietic stem cell transplantation (allo-HSCT).</p><p><b>METHODS</b>Six ALL patients who received sex-mismatched allo-HSCT and manifested extravisceral lymphadenectasis or local lump were investigated. The sex chromosome chimeric status in tumor tissues and bone marrows (BM) were monitored by FISH, and EBV-RNA in the tumor tissues were detected by in situ hybridization (ISH).</p><p><b>RESULTS</b>The sex chromosomes in BM of all 6 patients were 100% donor-derived. Among the sex chromosome chimeric status of tumor tissues, three patients were mainly recipient-derived, and the percentage of sex chromosomes derived from recipients were 100%, 100% and 98.0%, respectively, and then they were diagnosed leukemic extramedullary relapse. The other 3 patients were donor-derived, the percentage was 98.5%, 96.0% and 91.5%, respectively, and were diagnosed PTLD. EBV-RNA and latent membrane protein (LMP-1) were positive in 2 patients with PTLD and negative in the other 4 patients. One patient with extramedullary relapse obtained partial remission, one with PTLD gained complete remission, and the others died eventually after therapy.</p><p><b>CONCLUSION</b>Monitoring the sex chromosome chimeric status by FISH is an effective method to distinguish leukemic extramedullary relapse from PTLD in ALL received sex-mismatched donor HSCT.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expression of T cell receptor (TCR) Vgamma genes in patients with graft-versus-host disease (GVHD) after allogenic hematopoietic stem cell transplantation (allo-HSCT).</p><p><b>METHODS</b>The expression levels of the TCR Vgamma I-III genes in peripheral blood mononuclear cells (PBMNCs) from 18 patients with GVHD following allo-HSCT were determined using real-time fluorescence quantitative PCR, with 12 healthy individuals serving as the normal controls.</p><p><b>RESULTS</b>The expression level of TCR Vgamma II gene in the PBMNCs from patients with GVHD was significant lower than that in the normal controls. The expression patterns of TCR Vgamma I-III subfamilies also underwent alterations in patients with GVHD, and the expression level of TCR Vgamma II gene was significantly lower than that of TCR Vgamma I gene or TCR Vgamma III gene.</p><p><b>CONCLUSION</b>The low expression of TCR Vgamma II subfamily might be related to the pathogenesis of GVHD in patients receiving allo-HSCT.</p>
